1. Yes, you can use straws with a Basic Upgrade! You can pull straws from your own stallions or you can buy them from others to use! Your only limitation would be if a stallion has closed genes, in which case you would need a license to breed (which you can get with RL money or through the Lab some times).

2. Yes, you can use straws on either mares that you own or you can use "brood mares" that others offer publicly! So, if you have a good number of breedable mares and depending on how many straws you have, then you can get way more then just one foal from the same stallion! You can't use eggs from mares or make embryos with only a Basic Upgrade.
