I'm hoping someone can help me out with this boy.
I'd like to make him homozygous satin on strand one.

Hmmmm. I think you may be getting confused with the primary/secondary DNA. 1 copy of the rare gene will on both strands of DNA. So it will look like 2 copies. But 2 actual copies on this horse will be 4 copies total.
